Request shRNA Clones

The Hannon-Elledge human retroviral shRNA collection is available to all Duke investigator. Follow Clone Request Instructions (PDF file) as outlined below, and enter this information into online shRNA Clone Request System. Individual clones are available at a cost of $39.74 per clone.

To request a clone or clones:

1. Determine correct RefSeq accession number for target gene: http://www.ncbi.nlm.nih.gov/RefSeq/
RefSeq accession number should contain NM_ or XM_ prefix.

 

Hints:- Append 'AND "homo sapiens" [organism]' to search term to limit query to human RefSeq IDs. For example: c-myc AND "homo sapiens" [organism].
- To narrow down a large list of potential RefSeq IDs, select 'Limit' tab from query results page, and experiment with limiting query fields to 'Gene Name', 'Protein Name', 'mRNA', etc.

2. Query Open Biosystems database to identify appropriate CSHL Retroviral (Hannon-Elledge) shRNA' clone(s): Open Biosystems Query
Enter RefSeq # (alternatively, use keyword, gene symbol, or clone ID) into Query tool. CSHL (Hannon-Elledge) human retroviral shRNA clones will be listed, if available.

 

3. 'View' details of each clone and record Clone ID (v2HS_#).
*It is highly recommended to request and characterize all available clones targeting your gene of interest.

*NOTE: The Duke RNAi Facility has the human retroviral shRNA library through library release 1.15 - requesting clones in library release 1.16 or later from the shRNA Clone Request System will result in 'ID not valid' error message.

4. Login to shRNA Clone Request System (using NetID) and request desired clones. Confirm clone request list with information recorded above prior to completing request.

5. You will be notified when request is complete and bacterial stocks are available for pick-up from 215 Jones Bldg.
